The Hydragel 5 Proteinure Investigation for Renal Disease Classification
TEST PRESENTATION
Protein electrophoresis is a well-established technique for proteinuria screening. The HYDRAGEL 5 PROTEINURIE kit separates urine proteins according to their molecular weight on the HYDRASYS instrument.
The semi-automated HYDRASYS instrument allows migration and staining of the agarose gels, in order to obtain protein profiles for qualitative analysis. The resulting electrophoretic pattern is visually inspected and compared to molecular weight protein markers to characterize proteinuria.
This test allows the screening of the Bence Jones protein.
TEST PRINCIPLE
HYDRAGEL 5 PROTEINURIE separates proteins according to their molecular weight in a sieving agarose media after their treatment by an anionic detergent (SDS: Sodium Dodecyl Sulfate). The procedural parameters and sensitivity of the acid violet stain allows the detection of proteins in neat urines; no pre-analytical concentration required.
On HYDRAGEL 5 PROTEINURIE gels, the individual proteins of tubular origin (i.e., molecular weight < 65 – 70 kDa) and of glomerular origin (i.e., > 65 – 70 kDa) are clearly separated. Therefore, proteinuria can be characterized and classified according to the proteins’ molecular weight (e.g., glomerular, tubular and mixed).
The proteins are separated according to their molecular weight. The most current fractions that can be visualized are:
- ß2 Microglobulin (12 kDa)
- Lysozyme (15 kDa)
- RBP (21 kDa)
- Free Light Chains (≈25 kDa)
- α1 Microprotein (33 kDa)
- Dimers of Free Light Chains (≈50 kDa)
- Transferrin (80 kDa)
- IgG/IgA (150-160 kDa)
- Haptoglobins
- α2 Macroglobulin/IgM (800-900 kDa)
PERFORMANCES
Sensitivity:
- The detection level was determined with albumin and lysozyme solutions giving discernible bands at 15 mg/L.
