CD31, also known as Platelet Endothelial Cell Adhesion Molecule-1 (PECAM-1), is a member of the immunoglobulin (Ig) superfamily. CD31 is primarily concentrated at the lateral junctions of endothelial cells. It is also expressed on the surface of many types of cells, including platelets, monocytes, neutrophils, T- and B-cells, and natural killer cells.

Mature CD31 is composed of an extracellular domain comprising of six Ig-like domains (574 aa), a transmembrane segment (19 aa), and a cytoplasmic tail (118 aa). It has a molecular mass of 130 kD and is differentially glycosylated.

CD31 plays an important role in cell-cell interactions via either homophilic or heterophilic binding. A number of heterophilic binding partners have been reported, including glycosaminoglycans, integrin αvβ3, CD177, and CD38. CD31 functions as a negative regulator in immune cell activation and migration. CD31 is shed from platelets and controls platelet activation and aggregation. CD31 is also involved in regulating apoptosis by transmitting detachment or repulsion signals to viable or dying cells to phagocytes. Due to its wide expression and diverse functions, CD31 is involved in many diseases, including cancer, inflammation, atherosclerosis, and neovascularization. Human CD31 shares 64% and 61% aa identity with mouse and rat, respectively.
