Lymphopure™ – Reliable isolation without the cost

Lymphopure™

Lymphopure™ is a ready-to-use density gradient medium recommended for the isolation of mononuclear cells from peripheral blood and a cost-effective alternative to Ficoll Paque™ and Lymphoprep™. This endotoxin-filtered product from Biolegend has been used widely in research for over 40 years to study a wide array of immune cells including lymphocytes and granulocytes. Lymphopure™ takes advantage of differences in densities between different immune cells and the density gradient medium, with peripheral blood mononuclear cells (PBMCs) separating into the aqueous layer and Granulocytes and red blood cells sedimenting to the bottom due to their higher density.

Diagram showing whole blood cell separation using Lymphopure™. Blood is diluted with PBS and carefully layered over the medium in a conical tube, to avoid disturbing the interphase. Following centrifugation, at the appropriate speed with no braking, well-defined phases can be observed: 1-plasma, 2-interphase with mononuclear cells, 3-centrifugation medium, and 4-erythrocytes and granulocytes. Cells of interest can be aspirated from the interphase.

Benefits of using Lymphopure™

  1. High yield and purity: Lymphopure enables efficient separation of lymphocytes from other cell types present in a heterogeneous sample, such as red blood cells, granulocytes, and platelets. It offers high cell recovery rates and can yield a highly purified lymphocyte population.
  2. Simplicity and speed: The density gradient separation using Lymphopure is relatively straightforward and can be performed quickly. It eliminates the need for complex, time-consuming techniques such as red blood cell lysis or multiple centrifugation steps.
  3. Gentle separation: Lymphopure provides a gentle separation method that minimizes cellular stress and preserves cell viability. The low shear forces during centrifugation help maintain the integrity and functionality of the isolated lymphocytes, making them suitable for downstream applications.
  4. Scalability: Lymphopure is available in different volumes, allowing scalability for processing large or small sample volumes. It is compatible with standard laboratory equipment, making it easy to incorporate into various workflows.
  5. Consistency and reproducibility: Lymphopure is a well-characterized and standardized density gradient medium, ensuring consistent and reproducible results across different experiments or research studies. This reliability is crucial for comparing and interpreting data obtained from different sources.
  6. Versatility: Apart from lymphocyte isolation, Lymphopure can also be used for the separation of mononuclear cells from other tissue sources, such as bone marrow, spleen, or lymph nodes. Its versatility makes it a valuable tool in various immunology and cell biology research applications.
